Δημιούργησαν υγιή ωάρια ποντικών από βλαστικά κύτταρα

Ιάπωνες επιστήμονες δημιούργησαν στο εργαστήριό τους βιώσιμα ωάρια από βλαστικά κύτταρα, που είχαν ληφθεί από το δέρμα ποντικών και στη συνέχεια τα γονιμοποίησαν με εξωσωματική γονιμοποίηση παράγοντας τελικά υγιή φυσιολογικά ποντικάκια, τα οποία μάλιστα, σε επόμενο στάδιο, γέννησαν τους δικούς τους υγιείς απογόνους.

Το εντυπωσιακό επίτευγμα, που χαιρετίστηκε ως επιστημονικό ορόσημο στο πεδίο της τεχνητής αναπαραγωγής, αποτελεί σημαντικό βήμα που μελλοντικά μπορεί να επιτρέψει σε στείρες γυναίκες με προβληματικές ωοθήκες να παράγουν ωάρια και να αποκτήσουν παιδί, το οποίο θα έχει γενετική σχέση μαζί τους, καθώς θα μοιράζεται το μητρικό DNA. Ακόμα, θα μπορούσε να επιτρέψει στις γυναίκες να αποκτήσουν παιδί και μετά την εμμηνόπαυση, εξηγεί το ΑΜΠΕ.

Οι ερευνητές, με επικεφαλής τον Κατασουχίκο Χαγιάσι του πανεπιστημίου του Κιότο, που έκαναν τη δημοσίευση στο περιοδικό «Science» κατόρθωσαν για πρώτη φορά να δημιουργήσουν υγιή ωάρια ενός θηλαστικού, ικανά να παράγουν φυσιολογικούς απογόνους. Για να το πετύχουν αυτό, αξιοποίησαν την τεχνολογία των βλαστικών κυττάρων, μέσω της οποίας κοινά εξειδικευμένα κύτταρα (εν προκειμένω από το έμβρυο και το δέρμα ποντικών) μετατράπηκαν σε βλαστικά και αυτά, με τη σειρά τους, μέσω της κατάλληλης γενετικής τροποποίησης, προγραμματίστηκαν να γίνουν ωάρια που μετά ήταν δυνατό να γονιμοποιηθούν με σπέρμα. Τέλος, τα γονιμοποιημένα ωάρια εμφυτεύτηκαν στη μήτρα μιας παρένθετης μητέρας που γέννησε φυσιολογικά.

H αποδοτικότητα της μεθόδου είναι σχετικά χαμηλή. Πέντε ποντικάκια γεννήθηκαν από 127 εμφυτευμένα γονιμοποιημένα ωάρια (ποσοστό επιτυχίας περίπου 4%). Πολλά από τα έμβρυα όμως δεν αναπτύχθηκαν με φυσιολογικό τρόπο, χωρίς οι επιστήμονες να γνωρίζουν την αιτία.

Αν και υπάρχουν ακόμα πολλοί τεχνικοί και ηθικοί φραγμοί που πρέπει να ξεπεραστούν προτού η τεχνική εφαρμοστεί και στους ανθρώπους, οι Ιάπωνες επιστήμονες εμφανίζονται βέβαιοι ότι κάποια μέρα θα βοηθήσει πολλές γυναίκες να αποκτήσουν παιδί. Πάντως ο Χαγιάσι παραδέχτηκε ότι η εφαρμογή της τεχνικής στους ανθρώπους παραμένει μια μακρινή προοπτική, καθώς, μεταξύ άλλων, η κατανόηση των επιστημόνων για την ανάπτυξη του ανθρώπινου ωαρίου παραμένει ακόμα ανεπαρκής.

Η ίδια ερευνητική ομάδα είχε πέρυσι ανακοινώσει ότι είχε κάνει κάτι ανάλογο με αρσενικά ποντίκια, παράγοντας βιώσιμα σπερματικά κύτταρα επίσης με την μετατροπή κυττάρων του δέρματος σε βλαστικά, τα οποία στη συνέχεια μετατράπηκαν σε σπερματοζωάρια.

Άλλοι επιστήμονες εξήραν το έργο των Ιαπώνων συναδέλφων τους, όμως επεσήμαναν ότι τώρα θα πρέπει να κάνουν ένα ανάλογο πείραμα με δερματικά κύτταρα όχι εμβρύων αλλά ενηλίκων ποντικών, κάτι που, αν πετύχει, θα ανοίξει το δρόμο για δοκιμές και στους ανθρώπους, καθώς θα αρθεί η βασική ηθική αντίρρηση για χρησιμοποίηση ιστών από ανθρώπινα έμβρυα. Ακόμα, θα πρέπει να διασφαλιστεί η υγεία των απογόνων από τέτοια ωάρια σε βάθος χρόνου.

Deafness Cure Step Closer With Stem Cells

A cure for a common form of deafness known as auditory neuropathy is a step closer, after researchers from the University of Sheffield in the UK used human embryonic stem cells to repair a similar type of hearing loss in gerbils.
Project leader and stem-cell biologist Marcelo Rivolta and colleagues report their work in the 12 September online issue of Nature.

Many of the 275 million people worldwide with moderate-to-profound hearing loss have it because of a faulty link between the inner ear and the brain.

Using gerbils and human embryonicstem cells, Rivolta and colleagues describe how they repaired an important part of that link: the auditory nerve.

"We have the proof of concept that we can use human embryonic stem cells to repair the damaged ear," Rivolta told Nature News.

As well as proving that stem cells can repair damaged hearing, the researchers hope the breakthrough will lead to new treatments.

"More work needs to be done, but now we know it's possible," said Rivolta.

Scientists aim to start stem cell therapy of Alzheimer’s by 2016

A team of Korean scientists are striving to develop a stem cell therapy for Alzheimer’s disease by 2016 based on their latest discovery of how to grapple with the degenerative illness.

The team, led by professor Suh Yoo-hun at Seoul National University and RNL Stem Cell Research Institute head Ra Jeong-chan, said that their findings will open a new door to conquering Alzheimer’s disease.

``When we started experiments of injecting fat-derived human adult stem cells into mice with Alzheimer’s disease three years ago, we were skeptical. But we were surprised to learn that they substantially improved,’’ Suh said in a press conference.

``As far as cognitive functions are concerned, the mice nearly recovered to normal. In addition, we discovered that the adult stem cells have preventive effects on potential sufferers of Alzheimer’s.’’

Encouraged by the positive results of the animal tests, Suh’s team applied for an approval on clinical tests on human patients.

Alzheimer’s disease, which is typically accompanied by cognitive dysfunction and memory loss, has been touted one of the major maladies in fast-aging societies like Korea.

Currently, around half a million suffer from the most common form of dementia in Korea and the number is expected to double by the end of the second decade of the new millennium.

It is the sixth-leading cause of death and affects one in every eight people, which is more than breast cancer. As of 2010, there were 35.6 million people diagnosed with the disease in the world.

``Another positive part of the new-found therapy is that it does not involve any ethical issues or immune response problems because adult stem cells come from patients themselves,’’ Suh said.

``Fat-derived adult stem cells would not work on late-stage patients. But it may be used in a combined therapy of drug and stem cell for early-stage patients or potential sufferers.’’

Ra is very optimistic that the therapeutic effects of human stem cells can be applied for actual use in three years.

``Clinical tests would start this year at the earliest and would be completed within three years,’’ said Ra who founded and run RNL Bio, one of the top biotechnology start-ups in Korea.

``The year of 2016 will be remembered as the time when Alzheimer’s disease treatment effectively starts.’’

Ra said that the treatment is very stable because human administration of the kind of adult stem cell used in the experiment has been established in multiple articles and government-approved clinical trials.

``Injection of adult stem cells in bulk has been carried out several times and we found no abnormal aspects of development of cancers,’’ he said.

The results of their study were featured at Plos One, a peer-reviewed scientific journal, this week.

Stem cells in cord blood show promise for treating bowel disease

Washington, September 23 (ANI): Researchers have found a special population of stem cells in cord blood that has the innate ability to migrate to the intestine and contribute to the cell population there, suggesting the cells' potential to treat inflammatory bowel disease (IBD).
These cells are involved in the formation of blood vessels and may prove to be a tool for improving the vessel abnormalities found in IBD, said lead author Graca Almeida-Porada, M.D., Ph.D., a professor at Wake Forest Baptist Medical Center's Institute for Regenerative Medicine.
IBD, which is characterized by frequent diarrhea and abdominal pain, actually refers to two conditions - ulcerative colitis and Crohn's disease - in which the intestines become red and swollen and develop ulcers.
With IBD, blood vessels in the intestine leak and contribute to inflammation.
While there is currently no cure for IBD, there are drug therapies aimed at reducing inflammation and preventing the immune response. However, these therapies aren't always effective. The long-term aim of the research is to develop an injectable cell therapy to induce tissue recovery.
The work, performed while Almeida-Porada was at the University of Nevada, also involved colleagues from Indiana University School of Medicine.
The researchers studied a special population of cells, known as endothelial colony-forming cells, found in cord blood, bone marrow and circulating blood. The finding in 1997 that the cells can contribute to blood vessel formation in adults, not just embryos, initiated the notion of using them for therapy. Studies in humans have validated the ability of these cells to improve reduced blood flow to the limbs and to treat heart diseases.
However, there have been few studies to explore the inherent biologic ability of these cells to home to different organs and contribute to tissue-specific cell populations. Evaluating their potential to migrate to the intestine was an obvious choice, said Almeida-Porada, because dysfunctional blood vessels are a hallmark of IBD..............

Pioneering plan for stem cell hip replacements

SCOTS researchers have revealed plans to create a revolutionary new hip implant that will use the latest stem cell technology to allow patients to grow their own bone, removing the need for regular replacement surgery.
The prosthetic implants are being developed by scientists at Glasgow University in collaboration with orthopaedic surgeons at the city's Southern General Hospital and they hope to have a prototype ready within 10 years.
The move follows a breakthrough last year by a team at the university's Institute of Molecular, Cell and Systems Biology when they succeeded in creating a special plastic surface capable of controlling what stem cells become – a technique that was previously impossible.

The plastic is covered in tiny pits 120 nanometres across which, when stem cells are placed on the surface, encourages them to grow and spread across the pits in a way which ensures they differentiate into therapeutically useful cells.
While the implant itself will be made of an advanced polymer, already commonly used in spinal and other orthopaedic procedures, it will be coated for the first time in the university's unique plastic. As a result, once inserted into a patient's bone marrow – a rich source of mesenchymal stem cells – these can be made to divide and differentiate into fresh bone cells, creating a much stronger and more durable implant.
Dr Matthew Dalby, a specialist in bone tissue engineering at the institute, said: "By covering the implant in this surface, we can ensure the mesenchymal stem cells differentiate into the bone cells.
"This will help the implant site repair itself much more effectively than has ever been possible before and could well mean implants will last for the rest of patient's life.
"People are living longer – long enough, in fact, that we're outliving the usefulness of some of our body parts. Our new implant could be the solution to the expensive and painful follow-up surgeries which conventional implants require."
Currently, implants are commonly made from materials such as polyethylene, stainless steel, titanium or ceramic and tend to require replacement every 15 to 20 years.
In hip replacement surgery, the head of the thigh bone is removed and replaced with an implant which is held in place by a rod fixed inside the marrow along the length of the bone.
When traditional implants are fixed into bone marrow, the marrow's stem cells do not receive messages from the body to differentiate into bone cells.
Instead they usually differentiate into a build-up of soft tissue which, combined with the natural loss of bone density that occurs as people age, can weaken the bond between the implant and the body.
Safety concerns over metal hip implants were also raised earlier this year when it emerged thousands of Scots patients could be at risk from a build-up of toxic debris from DePuy ASR implants, which were first used on the NHS in 2003.
They were recalled last year after manufacturers found some were leaking cobalt and chromium into the blood, which has been linked with extreme pain, non-cancerous tumours and muscle wastage

ΘΕΡΑΠΕΙΑ ΕΓΚΕΦΑΛΙΚΩΝ ΕΠΕΙΣΟΔΙΩΝ ΜΕ ΒΛΑΣΤΟΚΥΤΤΑΡΑ

Επιστήμονες από το ερευνητικό κέντρο Mercy Gilbert Medical Center ανακοίνωσαν την πρώτη χορήγηση  μεσεγχυματικών βλαστοκυττάρων σε ασθενή με εγκεφαλικό επεισόδιο.  Η χρήση βλαστοκυττάρων θα επεκταθεί σε άλλους 35 ασθενείς και θα γίνει σε τρία συνεργαζόμενα κέντρα στις ΗΠΑ.  Η μέθοδος ήδη έχει εφαρμογή στην Ιαπωνία σε ασθενείς με πρόσφατο εγκεφαλικό  επεισόδιο και με χρήση μεσεγχυματικών βλαστοκυττάρων του λιπώδους ιστού.

ΕΡΓΑΣΤΗΡΙΟ, ΟΠΟΥ Η ΕΙΣΤΗΜΟΝΙΚΗ ΦΑΝΤΑΣΙΑ ΒΑΔΙΖΕΙ ΠΡΟΣ ΤΗΝ ΠΡΑΓΜΑΤΙΚΟΤΗΤΑ

Το εργαστήριο Αναγεννητικής Ιατρικής Wake Forest Institute for Regenerating Medicine των ΗΠΑ κάνει πράξη έργα επιστημονικής φαντασίας. Είναι το πρώτο εργαστήριο στον κόσμο που δημιούργησε ανθρώπινη ουρήθρα και  την μέθοδο θα την χρησιμοποιήσει για να βοηθήσει  5 παιδιά τα οποία υποφέρουν από κακώσεις της ουρήθρας  λόγω βαρέων τραυματισμών  της  πυέλου. Οι επιστήμονες πήραν ένα μικρό μέρος από την ουροδόχο κύστη και δημιούργησαν στο εργαστήριο ουρήθρα.  Δεδομένου ότι η ουρήθρα αποτελεί συνεχεία της ουροδόχου κύστης και έχει την ίδια ιστολογική κατασκευή  η δημιουργία της θεωρητικά δεν θα ήταν απίθανη. Στο εν λόγω εργαστήριο μια λογική σκέψη έγινε μεγάλη ιατρική εφαρμογή.
Ανάλογη μέθοδο έχουν εφαρμόσει στο ίδιο εργαστήριο δημιουργώντας πτερύγια αυτιών, τα οποία χορήγησαν σε τραυματισμένα πειραματόζωα.
Επίσης στο ίδιο εργαστήριο έχουν αναπτυχθεί μυς με καλλιέργεια μυϊκών κυττάρων,  που λαμβάνονται από βιοψία. Μέχρι σήμερα η καλλιέργεια και ο πολλαπλασιασμός των μυϊκών κυττάρων στο εργαστήριο δεν ήταν εφικτός.  Έτσι ο τραυματισμός μυός συνοδεύεται  από ελλείμματα και ουλές.  Σήμερα αναμένεται οι μυς που θα αναπτύσσονται στο εργαστήριο αυτό να εμφυτεύονται στις περιοχές των μυϊκών ελλειμμάτων,  στο πρόσωπο στις περιπτώσεις παράλυσης του προσωπικού νεύρου, στις νευροπάθειες και στην αποκατάσταση συγγενών ανωμαλιών του προσώπου , όπως του λυκοστόματος και του λαγώχειλους.   Δεν αποκλείεται βέβαια των μεθόδων αυτών να κάνουν χρήση και οι αθλητές για να βελτιώσουν τις επιδόσεις τους.
Όλες αυτές οι εφαρμογές ανοίγουν καινούργιους δρόμους στην αποκατάσταση μεγάλων βλαβών που μέχρι σήμερα ήταν αδιανόητη η αποκατάστασή τους.
