Baby was born in order to save brother with the use of umbilical cord stem cells

PARIS (AFP) – Doctors in France on Monday announced the country's first birth of a "saviour sibling," selected at the embryonic stage to be a close genetic match to save a brother or sister suffering from a fatal inherited disorder.
The baby was born at the Antoine Beclere Hospital in Clamart, in the suburbs of Paris, said doctors Rene Frydman and Arnold Munnich.
The child, born to parents of Turkish origin and named Umut-Talha (Turkish for "our hope"), was conceived through in-vitro fertilisation and was born on January 26 with a weight of 3.65 kilos (8.03 pounds), they said.
"He is in good health," Frydman told AFP.
The child's embryo was genetically selected to ensure he did not carry the gene for beta thalassemia, from which his siblings suffer, but was also a close enough match to provide treatment cells from umbilical cord blood, a rich source of stem cells.
Beta thalassemia produces an abnormal form of haemoglobin, the protein in red blood cells which carries oxygen around the body. It causes destruction of red blood cells which in turn leads to anaemia.
The world's first "saviour sibling" was Adam Nash, born in the United States in 2000.
The issue has been hedged with moral concerns about so-called designer babies and the morality of conceiving life to help a child with a genetic disorder.

ΕΓΚΡΙΘΗΚΕ ΚΛΙΝΙΚΗ ΜΕΛΕΤΗ ΜΕ ΤΗ ΧΡΗΣΗ ΠΟΛΥΔΥΝΑΜΩΝ ΒΛΑΣΤΟΚΥΤΤΑΡΩΝ ΓΙΑ ΤΗ ΘΕΡΑΠΕΙΑ ΤΗΣ ΠΕΡΙΦΕΡΕΙΑΚΗΣ ΑΓΓΕΙΑΚΗΣ ΝΟΣΟΥ.

Θεραπεία με βλαστικά κύτταρα αντιμετωπίζει την περιφερειακή αρτηριακή νόσο.           

 Η ισραηλινή εταιρία pluristem therapeutics  που δραστηριοποιείται στις θεραπείες με βλαστικά κύτταρα πέτυχε να πάρει την έγκριση των ΗΠΑ και της ΕΕ για να διεξάγει μια κλινική δοκιμή για την θεραπεία της νόσου των περιφερικών αρτηριών, η οποία αποτελεί  εκφυλιστική νόσο (νόσος φθοράς). Η ισραηλινή εταιρεία συμπλήρωσε την πρώτη φάση (Ι) της κλινικής δοκιμής και σύντομα θα ξεκινήσει την φάση ΙΙ/ΙΙΙ στην Ευρώπη και την Αμερική για την θεραπεία του τελικού σταδίου της νόσου περιφερικών αρτηριών. (critical limb ischemia).  Η νόσος αυτή συνίσταται σε απόφραξη αρτηριών, συνήθως στο κάτω άκρο η οποία προκαλεί πόνο, δυσκολία στο περπάτημα και εν τέλει οδηγεί σε ακρωτηριασμό του άκρου. Ο Chief executive Zami Aberman δήλωσε ότι η Pluristem θα διεξάγει μια κλινική μελέτη δεύτερης φάσης (ΙΙ) για να ελέγξει την θεραπεία σε ένα προγενέστερο/νωρίτερο/αρχικό στάδιο της νόσου προωθώντας έτσι την πρόληψη της νόσου. Η θεραπεία και η πρόληψη αφορά περίπου 20εκατ ασθενείς. Η μέθοδος αυτή είχε προταθεί εδώ και πολύ καιρό και ήταν ήδη γνωστό οτι η χρήση βλαστοκυττάρων στην ισχαιμία του κάτω άκρου βοήθησε να αποφευχθεί ο ακρωτηριασμός σε πολλές περιπτώσεις. «Θεωρούμε ότι το 2014 θα εγκριθεί και θα χρησιμοποιηθεί στην Ευρώπη και την Αμερική η θεραπευτική μέθοδος » δήλωσε ο Aberman. Η δεύτερη φάση της κλινικής δόκιμης θα περιλαμβάνει 150 ασθενείς και η φάση ΙΙ/ΙΙΙ θα διεξαχθεί σε όλη την Ευρώπη και τις ΗΠΑ και θα περιλαμβάνει 500 ασθενείς.

Scientists look to stem cells to mend broken hearts

Britain's leading heart charity launched a 50 million pound ($80 million) research project on Tuesday into the potential of stem cells to regenerate heart tissue and "mend broken hearts".
Scientists leading the work for the British Heart Foundation (BHF) said they hope that within the next decade they may have experimental drugs in development that would give certain kinds of cells in the heart the ability to regenerate tissue, repair damage and therefore combat heart failure.
The ability of heart tissue to regenerate already occurs in some animals, such as zebrafish, which can regrow portions of their own hearts if they are damaged.
At a briefing in London to launch a "mending broken hearts" fundraising campaign, scientists said research into stem cells and developmental biology may in future make this possible in people too.
"Scientifically, mending human hearts is an achievable goal and we really could make recovering from a heart attack as simple as getting over a broken leg," said Professor Peter Weissberg, medical director at the BHF.
Scientists in the United States reported last year that they had been able to turn structural heart cells into beating cells by identifying genes that, in a developing embryo, turn an immature cell into a beating heart cell or cardiomyocyte.
One of the British teams, led by Professor Paul Riley of the Institute of Child Health at University College London (UCL) has already found a natural protein, called thymosin beta 4, that plays a role in developing heart tissue.
He said his researchers had already had some success in using this protein to "wake up" cells known as epicardial cells in mice with damaged hearts.
"We hope to find similar molecules or drug-like compounds that might be able to stimulate these cells further," he told reporters at the briefing.
Another team of researchers at Imperial College London will be looking at a group of rare latent stem cells that can be harvested and then grown in the laboratory.
These cells are highly active in developing hearts and can grow into new functioning tissue, but something in them gets switched off soon after humans are born, meaning that the heart is no longer able to repair any damage, said Professor Michael Schneider, who leads this team.
His researchers will be trying to find ways of re-activating the cells in a controlled and safe way, so that they are able to repair damaged heart tissue but will not grow out of control.
"One strategy would be to give a drug that would activate this kind of process," he said, adding that this "requires more knowledge about what signals trigger these cells".
Weissberg said if the research was as successful as they hoped, it could one day reduce or even eliminate the need for heart transplants for patients whose hearts are damaged.

"Πιστόλι" Βλαστοκυττάρων Εναντίον Εγκαυμάτων !

Oμάδα ερευνητών στις ΗΠΑ κατασκεύασε ένα επαναστατικό εργαλείο με το οποίο γίνεται εύκολα, γρήγορα και ανώδυνα επούλωση εγκαυμάτων ακόμη και όταν αυτά είναι πολύ σοβαρά. Το εργαλείο ανέπτυξε ομάδα γιατρών με επικεφαλής τον καθηγητή Γιοργκ Γκέρλαχ του Ινστιτούτου Αναγεννητικής Ιατρικής του Πανεπιστημίου του Πίτσμπουργκ και στις δοκιμές που έγιναν είχε εξαιρετικά αποτελέσματα.

Το «πιστόλι δέρματος», όπως το βάφτισαν οι ειδικοί, είναι μια συσκευή που στην ουσία «πυροβολεί» την περιοχή η οποία έχει υποστεί τη ζημιά με βλαστικά κύτταρα. Η χρήση βλαστικών κυττάρων για τη δημιουργία δέρματος δεν είναι καινούργια. Και σήμερα οι ειδικοί αναπτύσσουν στο εργαστήριο δέρμα με βάση τα βλαστικά κύτταρα, το οποίο και τοποθετούν στην περιοχή των εγκαυμάτων. Ωστόσο αυτή η διαδικασία διαρκεί περίπου έναν μήνα, χρόνος που ειδικά σε περιπτώσεις ασθενών που έχουν υποστεί πολύ σοβαρά εγκαύματα είναι πολύτιμος, αφού πολλές φορές κάποιοι από αυτούς τελικά χάνουν τη μάχη με τη ζωή προτού γίνει η τοποθέτηση του νέου δέρματος.

Με τη νέα μέθοδο οι γιατροί αρχικά παίρνουν βλαστικά κύτταρα από ένα υγιές σημείο του δέρματος του ασθενούς και τα τοποθετούν σε διάλυμα νερού. Στη συνέχεια βάζουν το μείγμα στο «πιστόλι» και το ψεκάζουν επάνω στα εγκαύματα. Μετά τον ψεκασμό με τα βλαστοκύτταρα το έγκαυμα καλύπτεται με έναν επίδεσμο που περιέχει γλυκόζη, σάκχαρο, αμινοξέα, αντιβιοτικά και ηλεκτρολύτες, ώστε η πληγή να απολυμαίνεται και να θρέφεται ώσπου να αναλάβουν δράση τα βλαστοκύτταρα. Οι ειδικοί χαρακτηρίζουν το νέο εργαλείο το «ιερό δισκοπότηρο» του τομέα αποκατάστασης εγκαυμάτων.

Χαρακτηριστική είναι η περίπτωση του Μάθιου Ούραμ, αστυνομικού στην Πενσιλβάνια, ο οποίος υπέστη πολύ σοβαρά εγκαύματα στο πρόσωπο, στον λαιμό και στα χέρια κατά τη διάρκεια μπάρμπεκιου. Ο Ούραμ «ψεκάστηκε» από ειδικούς του νοσοκομείου στο οποίο κατέφυγε με το «πιστόλι» βλαστοκυττάρων και τρεις ημέρες αργότερα τα εγκαύματά του είχαν επουλωθεί πλήρως, όπως ο ίδιος δήλωσε σε ειδική εκπομπή που προέβαλε για αυτό το νέο ιατρικό εργαλείο του Νational Geographic Channel.

Induced Pluripotent stem (iPS) cells hold on to their past

Stem cells created from mature cells, called induced pluripotent stem cells, retain a distinct 'memory' of their former purpose that might limit their potential for therapeutic use.

Stem cells made from mature cells and rewound to an embryonic-like state retain a distinct "memory" of their past that might limit their potential for therapeutic use, scientists reported Wednesday in the journal Nature.

In a side-by-side comparison of these induced pluripotent stem cells and embryonic stem cells, researchers from the Salk Institute in San Diego found a consistent pattern of reprogramming errors — places where the iPS cells did not revert completely to an embryonic state.

"This study definitively demonstrates that there are differences between the two cell types," said Joseph Wu, a stem cell biologist at the Stanford University School of Medicine who was not involved in the research.

Knowing exactly what those differences are is a key step in developing ways to use iPS cells to treat diseases, experts said. Most researchers still consider embryonic stem cells to be the gold standard for regenerative medicine because they can grow into any type of cell in the body. But many scientists and policymakers would like to see treatments based on iPS cells because they can be made without destroying embryos. In addition, iPS cells can be custom-made for patients, ensuring a perfect genetic match.

To catalog the differences between the two cell types, the Salk team, led by molecular biologist Joseph Ecker, studied the epigenomes — chemical markers attached to DNA that regulate the way genes dial on and off — of 11 different cell lines.

They looked at 1.2 billion places in each genome where such chemical markers exist. The analysis was unusually rigorous — and therefore unusually revealing, Ecker said. Earlier studies examined representative regions in the genome, rather than the whole thing.

"Up to this point, people were looking through a keyhole," he said. "We're opening up the door."

For the most part, the contents of Ecker's metaphorical rooms looked alike. But when they zoomed in, inconsistencies emerged.

Large regions of the iPS epigenomes hadn't reverted to the embryonic state, but instead held on to the epigenetic memory of their tissue of origin. When the researchers used the iPS cells to create mature cells in the lab, this memory persisted.

The regions were clustered near telomeres and centromeres, structures that help direct how chromosomes divide.

"There is something about these regions that makes it harder to modulate the epigenome," Ecker said.

In some ways the iPS cells were different from one another, suggesting the reprogramming process itself might contribute to aberrations, he added.

That does not mean iPS cells can't be used in medicine, experts said. Improvements in technology could one day erase their epigenetic memory.

In addition, Wu said, scientists might find ways to harness that epigenetic memory to help treat disease. For example, if heart cells generated from iPS cells retain some of their cardiac characteristics, he suggested, they might be useful in therapies to treat heart disease."We need to do more research to see what exactly this means," he said.

New 3-D nanoscaffold could revolutionize human tissue engineering

Researchers from the University of Alabama at Birmingham's School of Engineering have created a three-dimensional electrospun scaffold on the nano scale that more effectively and efficiently facilitates cell and tissue growth in the laboratory.
Nanoscaffolds support the adhesion, growth and function of various cell types as they mature into specific tissues such as tendons, muscles and bones during tissue engineering. Yet, the traditional industry method for electrospinning creates densely packed sheet-like structures that prevent cells from penetrating the nanoscaffolds.
"Our three-dimensional electrospun nanoscaffolds better mimic nature and encourage cells to live longer and generate more viable, or functional, tissues," says Bryan Adam Blakeney, a recent graduate of the UAB Department of Biomedical Engineering  and a lead author on the study recently published online by the journal Biomaterials.
The discovery could lead to new applications for the regeneration of a damaged pancreas due to diabetes, among other tissues and body parts damaged by disease or other causes.
To achieve its three-dimensional scaffold, which it calls a FLUF - for focused, low-density, uncompressed nanofibrous mesh - the research team uses a spherical dish with a slight curvature similar to that of a home TV satellite during the electrospinning process.
"This allows the nanofibers that constitute the scaffold to intertwine and accumulate without becoming too tightly packed, which is the primary problem with flat, two-dimensional scaffolds," says another lead author Ajay Tambralli, a student in the UAB School of Medicine.
Blakeney and Tambralli, who were both UAB biomedical engineering undergraduates when they conducted the research, and their faculty mentor Ho-Wook Jun, Ph.D., assistant professor of biomedical engineering, worked two years to refine the process.
The researchers say the FLUF's more loosely packed nanofibers encourage cells to grow through the scaffold structure, creating the more stable formations that better mimic natural tissue.
"Think of it like a bowl of rocks: You pour a liquid into the bowl, and the liquid fills all the large gaps between the rocks," Blakeney said. "But with our 3-D nanoscaffold, our rocks, or the fibers that constitute the scaffold, are the size of sand particles. We've separated the grains of sand to create gaps, and the cells fill and penetrate the gaps between the sand grains to form tissues."
In addition to Blakeney, Tambralli and Jun, co-authors on the study include biomedical engineering graduate student trainees Joel Anderson and Adinarayana Andukuri; biomedical engineering graduate research assistant Dong-Jin Lim; and Derrick Dean, Ph.D., an associate professor in the UAB Department of Materials Science and Engineering.

Βλαστοκύτταρα από το Λίπος και χρήσεις τους στην Κοσμητική και Εξατομικευμένη Αναγεννητική Ιατρική

Τα  βλαστοκυτταρα σήμερα μπορούν να χρησιμοποιηθούν και στην αισθητική ιατρική, ακόμη και για τη φαλάκρα, την ανάπλαση του δέρματος  ή και την αύξηση του μαστού!
Σχετικά με την κοσμητική χρήση των βλαστικών και αναγεννητικών κυττάρων του λιπώδους ιστού, ο καθηγητής Βασίλης Πασπαλιάρης αναφέρει ότι ‘το λίπος του σώματός μας είναι μία από τις πιο προσιτές και πλούσιες πηγές σε βλαστοκύτταρα. Επιταχύνουν την ανάρρωση και αποκατάσταση της λειτουργίας των οργάνων του σώματός μας και ως εκ τούτου σήμερα αποτελούν χρήσιμα εργαλεία για τις νέες πιθανές μελλοντικές εφαρμογές στην εξατομικευμένη αναγεννητική ιατρική’.

Μία σημαντική εφαρμογή τους αφορά στην αποκατάσταση του μαστού ή την αύξηση του μεγέθους. Έχουν χρησιμοποιηθεί σε γυναίκες που υπέστησαν μαστεκτομή λόγω καρκίνου και τα αποτελέσματα ήταν πολύ καλά. Σε αυτές τις περιπτώσεις χρησιμοποιείται λίπος το οποίο είναι εμπλουτισμένο με βλαστοκύτταρα.
Πολύ αποτελεσματικά είναι όμως τα βλαστοκύτταρα και στις θεραπείες αντιγήρανσης και ομορφιάς. Χρησιμοποιούνται στο πρόσωπο για τη λείανση των ρυτίδων και την ανάπλαση της επιδερμίδας ώστε να φαίνεται περισσότερο νεανική, καθώς και για την αποκατάσταση των ουλών της ακμής.

Μία ακόμα κοσμητική εφαρμογή των βλαστοκυττάρων αφορά τη θεραπεία της φαλάκρας. Η έγχυσή τους με μεσοθεραπεία στην περιοχή του τριχωτού της κεφαλής βοηθά σημαντικά στην ανάπλαση της τρίχας. Μάλιστα οι νέες τρίχες είναι πιο υγιείς και δυνατές, ενώ βγαίνουν μαύρες ακόμα και εάν τα υπόλοιπα μαλλιά έχουν ασπρίσει!

Σήμερα η λήψη βλαστοκυττάρων για την αποκατάσταση της φθοράς του δέρματος γίνεται από το λιπώδη ιστό. Ο λιπώδης ιστός αποτελεί την πλουσιότερη πηγή λήψης βλαστοκυττάρων μεσεγχυματικού τύπου, τα οποία παράγουν τις σημαντικότερες πρωτεΐνες του δέρματος. Η λήψη των βλαστοκυττάρων από το λιπώδη ιστό γίνεται με λιποαναρρόφηση και η χρήση τους μπορεί να γίνει άμεσα ή να κρυοσυντηρηθούν και να χρησιμοποιηθούν στο μέλλον. Η χορήγησή τους γίνεται τοπικά στις ρυτίδες και για άμεσο αποτέλεσμα χορηγούνται συγχρόνως με μια ποσότητα λίπους ανάλογα με το βάθος της ρυτίδας.
Έτσι το λίπος δίνει ένα άμεσο αποτέλεσμα γεμίζοντας τη ρυτίδα και τα βλαστοκύτταρα αρχίζουν την παραγωγή των φυσιολογικών συστατικών του δέρματος τα οποία δίδουν ένα μόνιμο αποτέλεσμα. Το λίπος μετά από ορισμένο χρονικό διάστημα απορροφάται κι έτσι παραμένει η λειτουργική δραστηριότητα των βλαστοκυττάρων.

Τα μεσεγχυματικά κύτταρα σχηματίζουν το στηρικτικό σύστημα του σώματός μας αλλά και το σκελετό όλων των οργάνων επάνω στον οποίο στηρίζεται το λειτουργικό μέρος τους. Τα οστά, οι μυς, οι χόνδροι, τα αγγεία και οι τένοντες δημιουργούνται από τα μεσεγχυματικά κύτταρα. Με την πάροδο της ηλικίας η ανανέωση αυτών των οργάνων επιβραδύνεται με αποτέλεσμα την έκπτωση της λειτουργίας τους.

Χαρακτηριστικό παράδειγμα η εμφάνιση της οστεοπόρωσης, λόγω αδυναμίας των οστεοκυττάρων να παράγουν το σημαντικότερο συστατικό του σκελετού που είναι το κολλαγόνο. Σαν αποτέλεσμα τα ανόργανα άλατα που δίνουν τη σκληρότητα στα οστά μας δεν μπορούν να βρουν το φυσικό τους υπόστρωμα για να εναποτεθούν και γίνονται εύθραυστα.

Η χορήγηση μεσεγχυματικού τύπου βλαστοκυττάρων οδηγεί στον εμπλουτισμό των οργάνων με νεαρά κύτταρα τα οποία έχουν πλήρη λειτουργική δραστηριότητα, προκαλώντας έτσι μία γενική ανανέωση των ιστών του σώματός μας.  Όλοι οι ιστοί διαθέτουν βλαστοκύτταρα στο υπόστρωμά τους, το απόθεμά τους όμως εξαντλείται με την πάροδο της ηλικίας. Στον ενήλικα τα βλαστοκύτταρα παραμένουν αποθηκευμένα σε συγκεκριμένες θέσεις, εν υπνώσει και απαιτούνται ειδικές μέθοδοι συλλογής και διαχωρισμού. Ο λιπώδης ιστός διακρίνεται για το μεγάλο αριθμό των βλαστοκυττάρων που περιέχει. Σε συνδυασμό και με την εύκολη λήψη τους αποτελεί σήμερα την πολυτιμότερη πηγή λήψης βλαστοκυττάρων.

Πέρα όμως από την ανανέωση των οργάνων, τα βλαστοκύτταρα επιδρούν στο ανοσοποιητικό μας σύστημα, ασκώντας ρυθμιστική δράση και προστατεύοντάς το από υπερβολικές αντιδράσεις οι οποίες οδηγούν στην εμφάνιση των αυτοάνοσων παθήσεων. Οι κακώσεις του σώματος είτε είναι εμφανείς εξωτερικές, είτε εσωτερικές προκαλούν μεγάλες καταστροφές στα όργανα. Η χορήγηση μεσεγχυματικών κυττάρων οδηγεί σε ταχύτερη αποκατάσταση της λειτουργίας τους. Η χρήση τους ως μεταφορείς φαρμάκων και η κατευθυνόμενη χορήγησή τους σε όργανα αποτελεί μια νέα πολλά υποσχόμενη μέθοδο θεραπείας.
